Everyone so far has given you what you're looking for, now I want to give you the history of the song (the way I understand it anyway). "Rock Me Mama" was an unfinished song of Bob Dylan's. Old Crow Medicine Show took the chorus (which was written by Dylan) and added (or finished) the lyrics, and came up with the song "Wagon Wheel".

I think the term wagon wheel is referring to a certain juke box called the wagon whel which was made in the 1940s. There was a wooden wagon wheel on the front panel. Therefore, "Rock me like a wagon wheel" probably refers to rocking, moving, or dancing to the beat of the music the juke box plays.
